Microencapsulation is a technique that is widely used to overcome the problem of active substances in the free state by coating them in a polymer matrix. Several encapsulations have been carried out on active substances with different properties, and this gives different encapsulation (EE) values. Therefore, microencapsulation of active substances with different properties was carried out, namely ascorbic acid (AA) is acidic, nicotinamide (NT) is neutral, and theophylline (TF) is basic to further study the active properties of the EE value. Microencapsulation was carried out using chitosan (Cs) and alginate (Alg) polymers through complex coacervation and formulation using the Box-Behnken Design method. Statistical analysis of ANOVA was carried out to see the effect of the concentration of Cs and Alg, as well as the pH of the active substance on particle size, zeta potential, and EE. Furthermore, TEM analysis was carried out, in-vitro and physical drug evaluation. The microcapsules showed a particle size of about 0.31-5.06 µm. The concentration of Cs significantly affects the particle size. The average zeta potential value of microcapsules was -25.56 mV. The TTP crosslinker gave better results (-33.5 mV) than CaCl2 (-11.8 mV). The EE value of the microcapsules obtained was >90%, and the size of the active substance had a significant effect on the EE value, where AA microcapsules had an EE value greater than that of NT and TF microcapsules. The use of a combination of polymers forms a polyelectrolyte complex that is stronger so that it shows a higher EE value compared to the use of a single polymer. This is evidenced by the morphology of the microcapsules which have a spherical and solid structure. The microencapsulation technique with Cs and Alg polymers was able to increase the TF profile at pH 1.2 and NT media at pH 6.8 and showed good physical improvement in TF and NT microcapsules.
